该系列产品是专门为需要研发使用平流型诊断测试、或其它需要检测超低含量物质的医疗诊断和生物技术等科学研究而设计的系列产品。微粒具有蓝色或红色可见光,四种不同的荧光色,还具有四种不同的表面功能基团。微粒的粒径偏差系数(C.V.)低于5%,以确保在膜中的移动均一性。这些微粒具有0.4微米的公称直径,该尺度是大多数基于乳胶平流测试的最佳粒径尺寸,微粒的制备过程中未添加表面活性剂,因此勿需清洗,可直接使用。

平流测试开发用彩色和荧光微粒
Duke科学的微粒十几年来在世界上销量最佳的平流型诊断检测产品中得到了广泛应用。我们的彩色微粒中包含具有25%重量百分比的染料,大大高于其它同类产品。这种高信号强度的粒子在定性平流检测应用中提供了优异的测试灵敏度。近年来,定量型平流检测技术的出现,对于与其相匹配的彩色和荧光微粒的需求不断增加。除了提供高强度彩色微粒外,我们还提供具有同样微粒尺度及表面性能的荧光微粒。我们的荧光微粒发射光可以是绿光(激发光为接近紫外光和可见光波长)、红光和长波长发射光。荧光染料均匀的被包裹到聚合物微粒内,从而使微粒具有优良的光稳定性,并使染料完全地保留在微粒中。请见下表所示各种荧光微粒的具体技术数据。

具有四种不同表面功能基团的微粒
诊断测试产品的开发需要使用许多不同类型的抗体。为了满足所有包被方式的需求,我们开发了疏水(物理)吸附(疏水性的羧基和磺酸基功能团微粒)和共价偶联(反应性羧基修饰和醛基修饰微粒)两大类型的微粒。羧基修饰微粒的表面羧基密度是45-65 Å2/COOH,专门用于用碳二亚胺来进行的标准化学偶联方法。醛基修饰微粒的表面羧基密度是120-140 Å2/CHO,用于非常温和条件下的简单一步蛋白包被。Technical Note 13 描述了通过疏水物理吸附和共价偶联进行乳胶-蛋白包被的方法。

Trial Kits
Trial kits are available for research and development purposes. Each kit includes four bottles (5 mL each) of particle suspensions with one dye, and each of the 4 particle surface functionalities. Each row in the table represents one kit.

Each product includes a Material Safety Data Sheet (MSDS), a Certificate of Analysis, and Technical Note 13, which describes general methods for protein adsorption or coupling.

羧基修饰微粒
这些产品是通过共聚过程制备的,这就使得聚合物微粒表面带有反应性羧基负电荷。一般来说,羧基密度范围大约是10-125 Å2/COOH。微粒相对来说是亲水性的羧基可与水溶性的碳二亚胺试剂反应,然后与胺反应生成稳定的共价胺键。这种结合法在诊断测试上被广泛用于抗体吸附到微粒上。

醛基修饰微粒
醛基修饰微粒专门用于在非常温和的条件下(中性PH)一步法偶联具有胺基团的大分子。微粒表面的醛基密度是100-230 Å2/CHO,微粒表面静电荷是负电荷性表面。这些微粒与羧基修饰微粒在许多方面的应用相同,为蛋白质与微粒的结合提供了附加的途径。

羧基微粒(亲水性)
这些产品是均一的无表面活性剂的聚苯乙烯微粒,微粒表面的羧基起电荷稳定的作用。微粒的亲水性羧基靠近微粒的表面。尽管可以用来将物质共价偶联到粒子表面,但一般来说,它们用于物理吸附蛋白质。

磺酸基微粒(亲水性)
